In 2004, Keke made her debut in Barbershop 2: Back in Business. The following year, Keke signed a record deal with Atlantic Records. She was featured in Knights of the South Bronx (the television film) and starred in Second Time Around and ER (the television series). That same year, with the help of producer Ralph Farquhar, Keke starred in the movie role Keke and Jamal. 

In 2006, Keke had her acting breakthrough in the starring role in the film Akeelah and the Bee; for the role, she won an NAACP Image Award and Black Reel Award.
